Individual 2' Deoxyribonucleotide Phosphoramidites
Individual Deoxyribonucleoside Phosphoramidites
are produced from the corresponding 5'-monohosphates by dephosphorylation and chemical synthesis. Each nucleoside is derivatized as the 5'-O-(4, 4'-dimethoxytrityl) 3'-O-(2-cyanoethyl)-N-diisopropylphosphoramidite. dA and dC residues are further protected by 6-N- and 4-N-benzoyl groups respectively. dG residues are further protected by 2-N-isobutyryl groups. The products are supplied as a powder under argon gas.

NOTE:
Using a conventional oligonucleotide synthesizer without any modification to the software, the amount of each phosphoramidite required is as follows:
0.2 umole scale: 3.3 mg per each base required as labeled
1.0 umole scale: 16.5 mg per each base required as labeled
